Applications

Instructions for Use
**Phase 1: Tissue Collection & Transport**
- Collect tissue samples using provided instruments
- Place immediately in Tissue Transport Solution
- Transport at 4°C within 24-48 hours
**Phase 2: Tissue Processing**
- Rinse tissue with Tissue Rinsing Solution
- Mince into small fragments using sterile scissors
- Digest with Digestion Solution I for 30-60 minutes at 37°C
- Filter through cell strainer to obtain single cells or small clusters
**Phase 3: Organoid Establishment**
- Count viable cells using Trypan Blue staining
- Mix cells with pre-chilled Matrigel (on ice)
- Plate as domes in pre-warmed 24-well plate
- Incubate at 37°C for 10-15 minutes to solidify
- Overlay with complete Organoid Culture Medium
**Phase 4: Culture & Expansion**
- Change medium every 2-3 days
- Monitor organoid formation under microscope
- Passage when organoids reach optimal density (typically 7-14 days)
- Use Digestion Solution II for mechanical/enzymatic dissociation
**Phase 5: Cryopreservation**
- Harvest organoids at desired passage
- Suspend in Organoid Cryopreservation Solution
- Freeze gradually at -80°C, then transfer to liquid nitrogen
**Key Notes**: All procedures require aseptic technique. Maintain Matrigel on ice at all times.
