Product Description

The 'One-stop' Organoid Construction Kit developed is an all-in-one solution for organoid construction, integrating tissue sampling, organoid culturing, subculturing, cryopreservation, and resuscitation. It is characterized by its user-friendly nature and ready-to-use design, achieving a remarkably high success rate of over 90% in organoid construction. This product equips customers with all the necessary consumables and reagents throughout the entire experimental process, minimizing the time consumption on non-experimental procedures.

Applications

Human Gastric Organoid Construction Kit enables multiple applications in biomedical research and drug development:

1. **Disease Modeling**: Establish patient-derived organoid models that faithfully recapitulate tumor heterogeneity, histopathological features, and genomic profiles of original tissues for studying cancer biology, progression mechanisms, and therapeutic resistance.

2. **Drug Screening & Development**: High-throughput evaluation of drug efficacy, toxicity, and combination strategies using physiologically relevant 3D models. Supports testing of chemotherapeutics, targeted therapies, immunotherapies, and novel drug candidates.

3. **Precision Medicine**: Generate patient-specific organoid models for predicting individual treatment responses, enabling personalized therapeutic strategy selection and companion diagnostic development.

4. **Basic Research**: Study tissue development, stem cell biology, cell-cell interactions, organogenesis mechanisms, and disease pathogenesis in controlled 3D environments.

5. **Biobanking**: Establish and maintain organoid lines for long-term storage, distribution, and collaborative research through standardized cryopreservation protocols.

6. **Regenerative Medicine Research**: Explore potential applications of organoid-derived cells in tissue repair and transplantation approaches.

7. **Multi-omics Studies**: Compatible with downstream genomic, transcriptomic, proteomic, and metabolomic analyses for comprehensive molecular characterization.

Instructions for Use

**Complete Workflow Protocol**:

**Phase 1: Tissue Collection & Transport**
- Collect tissue samples using provided instruments
- Place immediately in Tissue Transport Solution
- Transport at 4°C within 24-48 hours

**Phase 2: Tissue Processing**
- Rinse tissue with Tissue Rinsing Solution
- Mince into small fragments using sterile scissors
- Digest with Digestion Solution I for 30-60 minutes at 37°C
- Filter through cell strainer to obtain single cells or small clusters

**Phase 3: Organoid Establishment**
- Count viable cells using Trypan Blue staining
- Mix cells with pre-chilled Matrigel (on ice)
- Plate as domes in pre-warmed 24-well plate
- Incubate at 37°C for 10-15 minutes to solidify
- Overlay with complete Organoid Culture Medium

**Phase 4: Culture & Expansion**
- Change medium every 2-3 days
- Monitor organoid formation under microscope
- Passage when organoids reach optimal density (typically 7-14 days)
- Use Digestion Solution II for mechanical/enzymatic dissociation

**Phase 5: Cryopreservation**
- Harvest organoids at desired passage
- Suspend in Organoid Cryopreservation Solution
- Freeze gradually at -80°C, then transfer to liquid nitrogen

**Key Notes**: All procedures require aseptic technique. Maintain Matrigel on ice at all times.
