The colon organoids of the crab-eating monkey (Macaca fascicularis, also known as cynomolgus monkey or crab-eating monkey) are derived from the stem cells of the colon of adult crab-eating monkeys.The colon tissue is located at the end of the digestive tract. Its main functions are water absorption and electrolyte balance. Its epithelium is composed of absorbent cells (enterocytes), goblet cells (goblet cells), enteroendocrine cells (EECs) and stem cells.Crab-eating monkeys are highly evolutionarily close to humans (genomic similarity is about 93%) and are an important model animal for pharmacokinetics and toxicology research.The colonoid organoids retain the ability of intestinal stem cells to self-renew and differentiate in multiple directions, and can reproduce the crypt-villi-like structure of the colon epithelium in the body.In 2021, Li et al. established the colon organoid of crab-eating monkeys for the first time and confirmed its application value in drug toxicity testing.The colon organoids of crab-eating monkeys can be cultured for a long time for at least 15 generations (about 75 days). The expression of stem cells and TA cell markers is higher than that of primary tissues, but the expression of enteroendocrine cell markers Chga and Chgb is lower in organoids.
