Cell Source and Tissue Characteristics:
Mouse tongue organoid differentiation medium is used to differentiate tongue epithelial stem cells into taste receptor cells (TRCs).The tongue organoids are derived from Lgr5+ stem cells from the mouse contour papillae (CVP) and foliate papillae (FoP).After differentiation, four taste cell types are produced: (1) Type I cells (glial-like cells)-expressing GLAST/NTPDase2; (2) Type II cells (receptor cells)-expressing PLCß2, TRPM5, GNAT3, responsible for sweetness, umami and bitter taste perception; (3) Type III cells (presynaptic cells)-expressing SNAP25, NCAM, with synaptic connections; (4) Type IV cells (precursor cells)-expressing Lgr5, SOX2.Wnt and Notch signals regulate the fate of taste cells.This differentiation system is suitable for the study of taste biology, taste disorders and taste phylogeny.
