Mouse pulmonary organoid differentiation medium is used to differentiate pulmonary bud tip progenitor cells or enlarged pulmonary organs into alveolar or airway lineages.Pulmonary bud tip progenitor cells (SOX2+/SOX9+/NKX2-1+) have two-way differentiation potential: (1) The direction of alveolar differentiation-the production of type II alveolar cells (AT2, SFTPC+/ABCA3+) and type I alveolar cells (AT1, AQP5+/HOPX+/PDPN+); (2) The direction of airway differentiation-the production of basal cells, epithelial cells, cup-shaped cells and ciliated cells.Alveolar differentiation requires Wnt activation and Notch inhibition, while airway differentiation requires precise regulation of SMAD signals.The differentiation system is suitable for the study of alveolar development, acute respiratory distress syndrome (ARDS), pulmonary fibrosis and repair of alveolar epithelial damage.
