Mouse liver organoid differentiation medium is used to differentiate liver progenitor cell organoids in the direction of mature liver cells (hepatocyte).Mouse liver organoids are derived from EpCAM+ bile duct cells or Lgr5+ liver stem cells, and maintain progenitor cell status under amplification conditions (expressing PROM1, AXIN2, SOX9, CD44, KRT19).After differentiation, the cells acquire the characteristics of mature liver cells, express albumin (Albumin/ALB), HNF4a, CYP450 family metabolic enzymes, α1-antitrypsin (AAT), etc., and have functions such as albumin secretion, cytochrome metabolic activity, and bile acid production.Notch signal inhibition is the key to driving liver cell differentiation.The differentiation system is suitable for drug metabolism research, toxicological testing and liver disease modeling.
