Mouse intestinal organoid differentiation medium is used to differentiate intestinal stem cell organoids into mature intestinal epithelial cell types.The mouse small intestine/colon organoids are derived from Lgr5+ intestinal stem cells, and the stem cell state is maintained under amplification conditions.After differentiation, four main intestinal epithelial cell types are produced: (1) enterocytes-expressing sucrose isomaltase (SI), alkaline phosphatase (ALPI), and Villin; (2) goblet cells-expressing Muc2 and TFF3; (3) enteroendocrine cells-expressing chromophilic granuloprotein A (ChgA/Chromogranin A); (4) Paneth cells (unique to the small intestine)-expressing chromophilic granuloprotein A (ChgA/Chromogranin A); (4) Paneth cells (unique to the small intestine)-expressing chromophilic granuloprotein A (ChgA/Chromogranin A).
