Mouse kidney organoids are derived from Six2+ nephron progenitor cells (NPCs) of embryonic kidney tissue (E12.5-E18.5), or obtained from iPSCs/ESCs targeted differentiation.The kidney is one of the most complex organs in the human body and contains more than 30 different cell types.Kidney organoids can form structures similar to glomeruli (podocytes), near-end tubules, loop of Henle, and far-end tubules.The culture of kidney organoids in mice involves simulating the key stages of kidney embryonic development: primitive streak → intermediate mesoderm→ metanephric mesenchyme → renal unit progenitor cells → renal unit (nephron).Kidney organoids are of great value for the study of kidney development, polycystic kidney disease (ADPKD), acute kidney injury, drug nephrotoxicity screening, and renal regenerative medicine.In 2024, Li et al. reported a YAP activation strategy based on mNPSR-v2 medium in Cell Stem Cell, which achieved long-term amplification of mouse and human renal unit progenitor cells.
