Cell Source and Tissue Characteristics:
Nasopharyngeal Carcinoma (NPC) is a malignant tumor that originated in the epithelial cells of the nasopharynx. It is closely related to Epstein-Barr Virus (EBV) infection and is highly common in southern China and Southeast Asia.NPC is divided into three WHO subtypes: keratinized squamous cell carcinoma (type I), non-keratinized differentiated cancer (type II), and non-keratinized undifferentiated cancer (type III).NPC organoids are derived from biopsy or surgical resection specimens of NPC patients, and all NPC organoids are EBER positive (EBV coding RNA in situ hybridization) and CK7 negative.Wang et al. (2022) established an organoid biobank containing 34 primary NPCs and 28 recurrent NPCs. The success rate of organoid establishment of primary NPCs was 47.06%, and that of recurrent NPCs was 81.25%.Relapsed NPC organoids have higher expression of stem cell markers (BMI-1, CD44, CD133).NPC organoids retain the EBV infection status and molecular heterogeneity of the primary tumor, and can be used for drug screening and personalized treatment research.
