Dissociation Enzyme Solution II

This product is designed for dissociation of various organoids and may also be used for selective tissue dissociation. It features high purity and excellent specificity, preventing potential cellular damage from contaminating enzymes present in crude trypsin extracts. Ideal for routine organoid passaging and subculture applications.

(1) Remove Enzyme Solution II from 2-8°C refrigerator and equilibrate at room temperature for 10 minutes before use. (2) Using a 1000 μL pipette tip pre-rinsed with Rinse Solution, add appropriate volume of Enzyme Solution II to the centrifuge tube containing organoids (refer to Table 1), resuspend organoids, and transfer organoid suspension to a 24-well plate. (3) Place the 24-well plate in a 37°C, 5% CO₂ incubator and digest for approximately 5-15 minutes. Every 5 minutes, pipette organoids 5-10 times using a pre-rinsed 1000 μL pipette tip. Monitor dissociation under microscope; when most cells appear as single cells or small clusters, terminate enzymatic digestion.
