Description

This product is suitable for dissociation of various tissues. It features high purity and excellent specificity, avoiding potential cellular damage from contaminating enzymes commonly found in crude trypsin preparations. Optimized for primary tissue dissociation to establish organoid cultures from fresh specimens.

Instructions

(1) Remove Dissociation Enzyme Solution I from -20°C freezer and thaw at room temperature before use. (2) Tissue dissociation: 1) Use ophthalmic scissors to cut tissue into pieces approximately 0.5-1 mm in diameter. 2) Transfer tissue fragments to a 15 mL centrifuge tube, centrifuge at 200×g for 5 minutes at 2-8°C to collect tissue. 3) After centrifugation, use a Pasteur pipette to carefully remove supernatant without disturbing the pellet. Based on remaining tissue volume, add corresponding volume of Enzyme Solution I using a 1000 μL pipette (for tissue pellet < 1 mL, add 5 mL; for 1-2 mL, add 10 mL). Resuspend using a pre-rinsed Pasteur pipette and transfer to a new 6 cm or 10 cm culture dish. Place in a 37°C, 5% CO₂ incubator for enzymatic dissociation. 4) During dissociation, remove the dish every 10 minutes, mix by pipetting 5-10 times using a pre-rinsed Pasteur pipette, and monitor dissociation progress under microscope.
