Cell Source and Tissue Characteristics:
Mouse hepatocellular carcinoma (HCC) organoids are derived from genetically engineered mouse models (GEMMs) or chemically induced liver cancer tissues, including: (1) genetically engineered models-such as MYC/TGFa double transgenic mice, TAK1 knock-out mice, PTEN knock-out mice, etc.; (2) Chemically induced models-diethylnitrosamine (DEN) induction, carbon tetrachloride (CCl4) combined with DEN induction, choline deficiency L-amino acid definition high-fat diet (CDAHFD) induced MASH-HCC model.HCC organoids retain the genetic mutations, histopathological characteristics and gene expression profiles of the primary tumor.The organoids are in a sac-like or solid form and express the HCC markers AFP, GPC3, and HNF4a, which can be used for drug screening and drug resistance mechanism research.This model is suitable for the study of the mechanism of HCC occurrence, drug development and personalized treatment.
