Cell Source and Tissue Characteristics:
Human lung cancer organoids are derived from surgical resection specimens or biopsy samples of lung cancer patients. Organoids of various histological subtypes such as pulmonary adenocarcinoma (LUAD), pulmonary squamous cell carcinoma (LUSC), large cell neuroendocrine carcinoma (LCNEC) and small cell lung cancer (SCLC) can be established.The organoids of lung cancer retain the histopathological characteristics, gene mutation profiles and drug response characteristics of the primary tumor.The main gene mutations include KRAS, EGFR, TP53, ALK rearrangement, ROS1 fusion, etc.The success rate of NSCLC organoid establishment reported by Sachs et al. (2019): surgical specimens were 88% (14/16) and metastatic biopsy specimens were 28% (5/18).Kim et al. (2019) reported a higher success rate (87%, 20/23) and established a live bank containing 80 lung cancer organoids.Lung cancer organoids can be used for personalized drug screening, targeted treatment response prediction, and drug resistance mechanism research.
