Cell Source and Tissue Characteristics:
Human normal liver organoid is derived from adult liver tissue bipolar stem cells (bipolar stem cells), these cells have the ability to differentiate into hepatocytes (hepatocytes) and cholangiocytes (cholangiocytes).Tissue sources include discarded tissue margins from liver transplantation, normal marginal tissue from liver resection surgery, or liver biopsy samples.Liver organoid forms a cystic structure in vitro and expresses liver cell markers (ALB, AAT, CYP3A4) and bile duct cell markers (KRT19, KRT7, SOX9).Huch et al. (2015) demonstrated for the first time that there are dual-potential liver stem cells in the adult liver that can be enlarged for a long time and maintain genomic stability. These stem cells can renew themselves in culture and differentiate into functional liver cells and bile duct cells.
