Human glioma organoids are derived from surgically resected glioblastoma (GBM) tumor tissue, representing WHO Grade IV malignant brain tumors. GBM is the most common and aggressive primary brain tumor, characterized by high cellular heterogeneity comprising glioma stem cells (GSCs), tumor cells, immune cells, and vascular endothelial cells. Key genetic features include IDH-wildtype status, variable MGMT promoter methylation, common EGFR amplification, and PTEN loss. Tumor tissue is optimally collected from tumor margin regions with minimal necrosis and surrounding normal brain tissue to maximize preservation of tumor cell heterogeneity and cell-cell interactions.
