Human Airway Organoids (Human Airway Organoids) are derived from human bronchial epithelial cells (HBECs) or human airway epithelial cells (HAECs), mainly based on bronchial-alveolar lavage fluid, surgically removed airway tissue or brushing samples.The airway organoids simulate the structure of the respiratory tract from the trachea to the bronchial tree, including basal cells (TP63+/KRT5+), multiciliated cells (FOXJ1+/acetylated tubulin+), secretory cup-shaped cells (MUC5AC+/MUC5B+) and CC10+Club cells (Club cells).As adult stem cells in the airway, basal cells have the potential for self-renewal and multi-directional differentiation.Airway organoids can be used to simulate diseases such as asthma, cystic fibrosis (CF), COPD, primary ciliary dyskinesia (PCD), and respiratory infections.Sachs et al. (2019) established a long-term amplified human tract organoid model, proving that it can maintain culture for more than 1 year and maintain a stable multi-lineage differentiation ability.
