Red Blood Cell (RBC) Lysis Buffer

This sterile-filtered lysis buffer is designed for efficient lysis of red blood cells from blood or tissue samples. Processed samples can be used for subsequent primary cell culture, cell fusion, nucleic acid or protein extraction, and various routine analytical assays.

(1) Store the product immediately upon receipt according to the recommended conditions. (2) Process fresh tissue through enzymatic digestion and disperse into a single-cell suspension by appropriate methods, then centrifuge and discard supernatant. (3) Add 3-5x cell pellet volume of RBC Lysis Buffer, gently pipette to mix, and lyse for 1-2 minutes. For example, if the cell pellet volume is 1 mL, add 3-5 mL of RBC Lysis Buffer. This step may be performed at room temperature or 2-8°C. (4) Centrifuge at 400-500×g for 5 minutes and discard the red supernatant. Centrifugation at 2-8°C yields better results. (5) If lysis is incomplete, repeat steps (3) and (4) once.
