Human pulmonary organoid differentiation medium is used to induce differentiation of pulmonary stem cells or pulmonary bud progenitor cells into functional alveolar and/or airway cell types.The direction of differentiation depends on the medium formula: the distal differentiation scheme (distal differentiation) produces alveolar organoids, including AT2 cells (SFTPC+/HTII-280+/ABCA3+) and AT1 cells (AQP5+/PDPN+/HOPX+); the proximal differentiation scheme (proximal differentiation) produces airway organoids, including basal cells, ciliated cells, and cup-shaped cells.The key to alveolar differentiation is to simulate the differentiation process of AT2 cells to AT1 cells, which is the core mechanism of alveolar regeneration and repair.Barkauskas et al. (2013) demonstrated for the first time that AEC2 is an alveolar stem cell that can self-renew and differentiate into AEC1.
