Vocal cord Leukoplakia (VCL) is one of the most common precancerous lesions of the larynx. It is a transitional lesion that progresses from benign vocal cord polyps (VCP) to laryngeal squamous cell carcinoma (LSCC).VCL is characterized by white plaques of the mucosal epithelium of the vocal cords, which are histologically manifested as abnormal hyperplasia to varying degrees.The molecular mechanism by which VCL develops into LSCC is not completely clear.VCL organoids are derived from surgical resection specimens of VCL patients, which can simulate the continuous process from benign lesions to malignant progress.Recent single-cell RNA sequencing studies have revealed that VCL has a "mixed microenvironment" with mixed VCP and LSCC characteristics, as well as signs of early immunosuppressive activity.LSCC organoids can be derived from primary tumors of the larynx, including glottis, supraglottis, and transglottis.VCL/LSCC organoids provide a unique platform for studying the molecular mechanisms of laryngeal cancer and developing early intervention strategies.
