Cell Source and Tissue Characteristics:
Human normal pancreatic organoid is derived from ductal epithelial stem cells of adult pancreatic tissue, mainly from surgically removed pancreatic tissue (such as the normal pancreatic area of pancreatic duodenal resection specimens) or organ donor pancreas.The pancreatic organoid forms a cystic structure in vitro, reproduces the histological characteristics of the pancreatic duct epithelium, and expresses duct markers (KRT19, KRT7, SOX9, CFTR), pancreatic progenitor cell markers (PDX1, NKX6.1) and endocrine markers.Normal pancreatic organoid can be further differentiated into endocrine-like cells expressing insulin (INS), glucagon (GCG), and somatostatin (SST).Compared with PDAC organoid, normal pancreatic organoid exhibits stronger Wnt dependence, is more regular in morphology, and has a higher degree of cystic fibrosis.
