Esophageal cancer organoid is mainly derived from esophageal squamous cell carcinoma (Esophageal Squamous Cell carcinoma, ESCC) and esophageal adenocarcinoma (Esophageal Adenocarcinoma, EAC).ESCC is the most common histological type, which mainly occurs in the upper middle esophagus and is closely related to smoking and alcohol consumption; EAC occurs in the lower esophagus and cardia, and is closely related to Barrett's esophagus and obesity.Esophageal cancer organoid retains the histological characteristics, gene mutation profile (TP53 mutations are the most common in ESCC) and drug response characteristics of the primary tumor.The accumulation of nuclear p53 protein in ESCC organoid is an important sign that distinguishes it from normal esophageal organoid.The study found that almost all ESCC organoid lines can be continuously passed down for more than 3 months, while normal esophageal organoid can only be maintained for about 30 days.
