Chronic obstructive pulmonary disease (COPD) is a chronic inflammatory disease characterized by persistent respiratory symptoms and irreversible airflow restriction, mainly caused by smoking and exposure to air pollutants.COPD pulmonary organoids are derived from airway biopsy (bronchi) or nasal mucosa tissue of COPD patients, and can also be established by exposing normal organoids to cigarette smoke extract (CSE).COPD organoids retain the characteristic pathological changes of the disease: goblet cell hyperplasia, increased MUC5AC expression, decreased cilia and decreased frequency of cilia pulsation.Chan et al. (2022) systematically established COPD nasal mucosa and bronchial organoids for the first time, and found that they showed disease characteristics consistent with the body, including secretory cell metaplasia and abnormal differentiation trajectories of airway epithelium.COPD organoids can be used to study host-pathogen interactions (such as SARS-CoV-2 and Pseudomonas aeruginosa infections), and it has been found that COPD organoids produce a stronger pro-inflammatory response to viral and bacterial exposure.
